Overview Accu Test HCV Chromatographic Immunoassay
The AccuTest HCV rapid chromatographic immunoassay is a qualitative screening test detecting antibodies targeting conserved core, NS3, NS4, and NS5 HCV proteins in human whole blood, serum, or plasma. Hepatitis C virus (HCV), a small, enveloped, positive-sense, single-stranded RNA virus, is notoriously difficult to isolate using conventional methods. However, genomic cloning enabled the development of serological assays employing recombinant antigens. The AccuTest HCV assay utilizes multiple recombinant protein antigens to enhance sensitivity and minimize cross-reactivity. This lateral flow immunoassay employs a double-antigen sandwich technique. Recombinant HCV antigen coats the test line; sample interaction with antigen-coated particles, followed by capillary migration, produces a colored line if antibodies are present. A control line confirms proper procedure. **Procedure:** Bring all components to room temperature. Add one drop (30µL) of sample and two drops (40µL) of buffer to the sample well. Read results within 15 minutes (strong positives may appear within 5 minutes); results after 15 minutes are invalid. **Interpretation:** A purple line at the 'T' (test) and 'C' (control) lines indicates a positive result (presence of HCV antibodies). A purple line only at 'C' is negative. Absence of both lines or lack of a 'C' line indicates an invalid test. **Storage and Safety:** Store at 4-30°C; unopened pouches may be stored at room temperature (≤30°C). How to Use Accu Test HCV Chromatographic Immunoassay:
Bring the test, sample, and/or control to room temperature prior to analysis. Extract a single test card from its packaging and lay it on a clean, level surface. Employ the provided dropper to dispense one drop of serum/plasma or whole blood (30µL) followed by two drops of buffer (40µL) directly into the sample well. Prevent spillage. Interpret the results within 15 minutes; a strongly positive reaction might appear within 5 minutes. Disregard any readings obtained after 15 minutes.
Safety Instruction of Accu Test HCV Chromatographic Immunoassay:
- AccuTest HCV requires refrigeration between 4°C and 30°C. Alternatively, unopened pouches may be kept at room temperature, provided the temperature remains below 30°C.
- Always check the product label before administration.
- Store in a location inaccessible to children.
